Abstract:
Polycomb-like proteins 1-3 (PCL1-3) are substoichiometric components of the Polycomb-repressive complex 2 (PRC2) that are essential for association of the complex with chromatin. However, it remains unclear why three proteins with such apparent functional redundancy exist in mammals. Here we characterize their divergent roles in both positively and negatively regulating cellular proliferation. We show that while PCL2 and PCL3 are E2F-regulated genes expressed in proliferating cells, PCL1 is a p53 target gene predominantly expressed in quiescent cells. Ectopic expression of any PCL protein recruits PRC2 to repress the INK4A gene; however, only PCL2 and PCL3 confer an INK4A-dependent proliferative advantage. Remarkably, PCL1 has evolved a PRC2- and chromatin-independent function to negatively regulate proliferation. We show that PCL1 binds to and stabilizes p53 to induce cellular quiescence. Abstract:
Optimising non-viral vectors for neuronal siRNA delivery presents a significant challenge. Here, we investigate a co-formulation, consisting of two amphiphilic cyclodextrins (CDs), one cationic and the other PEGylated, which were blended together for siRNA delivery to a neuronal cell culture model. Co-formulated CD-siRNA complexes were characterised in terms of size, charge and morphology. Stability in salt and serum was also examined. Uptake was determined by flow cytometry and toxicity was measured by MTT assay. Knockdown of a luciferase reporter gene was used as a measure of gene silencing efficiency. Incorporation of a PEGylated CD in the formulation had significant effects on the physical and biological properties of CD. siRNA complexes. Co-formulated complexes exhibited a lower surface charge and greater stability in a high salt environment. Abstract:
Cocrystal formation between nicotinamide and five fenamic acid derivative drugs (flufenamic acid, niflumic tolfenamic acid, mefenamic acid and meclofenamic acid) was investigated using solution-based and solid-state preparation methods. It was anticipated that the well-known acid-aromatic nitrogen heterosynthon would provide a sufficient driving force for cocrystallization. The experiments yielded cocrystals with four of the five acids. Although the structures of these molecules are similar, they showed marked differences in both the stability and the stoichiometry of the cocrystals. A detailed analysis of the structures and properties of both the starting materials and the cocrystals allows a tentative explanation of these differences, but it also shows that even though all four cocrystals utilize one of the most predictable supramolecular synthons (COOH center dot center dot center dot N), their structures and properties remain elusive to design.

Abstract:
Oxidation state (OS) is defined using ionic approximation of bonds. Two principal algorithms are outlined for OS determination in a chemical compound described by a Lewis formula or bond graph. Typical origins of ambiguous OS values are pointed out, and the relationship between OS and the d(n) electron configuration of transition metals is commented on.

Abstract:
We study the dynamics of a two-level quantum system interacting with an external environment that takes the form of an XY spin chain in the presence of an external magnetic field. While the presence of the bath itself can enhance the transition probability from the lower level to the upper level of the system, we show that this noise-assisted phenomenon is sensitive to a change of the quantum phase of the environment. The derivative of the transition probability displays a maximum in correspondence with the critical value of the applied field in the case of both isotropic and anisotropic chains.

Recent research in yeast and mammalian cells shows that gamma-H2AX facilitates post-replicational DNA repair by recruiting cohesin, a protein complex that holds sister chromatids together.

Abstract:
Early medieval settlement archaeology utterly dominates the record of excavations in Ireland, including settlement enclosures, complexes, landscapes and ecclesiastical sites (O'Sullivan, McCormick, Kerr & Harney 2009). For this reason, the INSTAR-funded Early Medieval Archaeology Project (EMAP) focused its research in 2009-2010, on dwellings and settlements (having previously provided a review of all early medieval archaeological excavations in Ireland). In 2009, EMAP first prepared a gazetteer of what we would regard as Ireland’s key early medieval settlements and dwelling excavations, largely based on a detailed review of the original EMAP database, 1930-2004; amounting to a final gazetteer (see Vol II) of 241 early medieval settlements revealed through archaeological excavation, 1930-2009. Background: Successful delivery of small interfering RNA (siRNA) to the lungs remains hampered by poor intracellular delivery, vector-mediated cytotoxicity, and an inability to withstand nebulization. Recently, a novel cyclodextrin (CD), SC12CDClickpropylamine, consisting of distinct lipophilic and cationic subunits, has been shown to transfect a number of cell types. However, the suitability of this vector for pulmonary siRNA delivery has not been assessed to date. To address this, a series of high-content analysis (HCA) and postnebulization assays were devised to determine the potential for CD-siRNA delivery to the lungs. Water-soluble 4-(dimethylamino) pyridine (DMAP) stabilized gold nanoparticles (DMAP-AuNP) were synthesized by ligand exchange and phase transfer (toluene/water). The DMAP-AuNPs are positively charged with the core diameter of 4 +/- 1 nm. Metallopolymer-gold nanocomposites were prepared by mixing gold nanoparticles and [Ru(bpy)(2)PVP(10)](ClO(4))(2), in water at different mole ratios: bpy is 2,2'-bipyridyl and PVP is poly (4-vinylpyridine). The photoluminescence emission intensity of the metallopolymer decreases with increasing AuNP loading and approximately 57% of the emission intensity is quenched when the Au NP:Ru mole ratio is 14.8 x 10(-2). The rate or homogeneous charge transfer through thin layers of the nanocomposite deposited oil glassy carbon electrodes increases with increasing nanoparticle loading. Abstract:
The dimeric complex [Mn-2(III)(Naphth-sao)(2)(Naphth-saoH)(2)(MeOH)(2)]center dot 4MeOH (1.4MeOH), acts as a simple model complex with which to examine the magneto-structural relationship in polymetallic, oxime-bridged Mn-III complexes. Dc magnetic susceptibility studies reveal that ferromagnetic exchange is mediated through the heavily twisted Mn-O-N-Mn moiety (J = + 1.24 cm(-1)) with magnetisation measurements at low temperatures and high fields suggesting significant anisotropy. Simulations of high field, high frequency EPR data reveal a single ion anisotropy, D-(MnIII) = -3.94 cm(-1). Theoretical studies on simplified model complexes of 1 reveal that calculated values of the exchange coupling and the anisotropy are in excellent agreement with experiment, with the weak ferromagnetism resulting from an accidental orthogonality between the Mn-N-O plane of the first MnIII ion and the Jahn-Teller axis of the second Mn-III ion.

Abstract:
We present a first principles static and dynamical study of the transition metal hydride series MH4L3 (M = Fe, Ru and Os; L = NH3, PH3 and PF3), with a view to arriving at an understanding of how the variation in the electronic properties of the metal sites and ligands can influence the dynamics of the resulting complexes. A broad range of behaviour was observed, encompassing stable classical minima (M = Os, L = NH3 and M = Ru, L = PH3) to stable eta(2)-H-2 non-classical minima (M = Fe, L = PF3 and M = Ru, L = PH3 or PF3), with the other structures exhibiting dynamical behaviour that spontaneously converted between the classical and non-classical states during the molecular dynamics simulations. The importance of a small L-axial-M-L-axial angle in stabilising the non-classical state is highlighted, as is a short eta(2)-H-2 center dot center dot center dot H-cis distance in non-classical complexes that spontaneously convert to the classical form. Abstract:
The synthesis, structural and magnetic characterisation of a family of homo- and heterometallic complexes constructed with the Schiff base ligands 2-iminomethyl-6-methoxy-phenol (L1H) and 2-imino-6-methoxy-phenol (L2H), are discussed. Members include the heterometallic tetranuclear complexes of general formula [Na2M2(X)(2)(L-1)(4)(Y)(2)] (where M = Fe-III, X = -OMe, Y = NO3- (1) and M = Ni-II, X = N-3(-) and Y = MeCN (2)), each possessing a butterfly-like topology. We also report the formation of the heterometallic molecular cage [Na3Ni2(L-1)(6)](ClO4) (3) whose metallic skeleton describes a [rare] trigonal bipyramid, the homometallic 1-D coordination polymer [Mn(L-1)(2)(Cl)](n) (4), and the tetranuclear cubane clusters [(Mn3MnIV)-Mn-III(O)(3)(OEt)(OAc)(3)(L-1)(3)] (5) and [Ni-4(mu(3)-OMe)(4)(L-2)(4)(MeOH)(4)] (6). Dc and ac magnetic susceptibility studies on complexes 5 and 6 reveal S = 9/2 and S = 4 spin ground states.

A family of homo-valent [Co(II)(7)(OH)(6)(L-1)(6)](NO3)(2)(1), [(MeOH)(2)subset of Co(II)(7)(OH)(6)(L-1)(6)](NO3)(2) (2) (where L1H = 2-iminomethyl-6-methoxyphenol) and hetero-valent [(NO3)(2)subset of Co(III)Co(II)(6)(OH)(6)(L-2)(6)]-(NO3)center dot 3MeCN (4) (where L2H = 2-iminophenyl-6-methoxyphenol) complexes possess metallic skeletons describing planar hexagonal discs. Their organic exteriors form double-bowl shaped topologies, and coupled with their 3-D connectivity, this results in the formation of molecular cavities in the solid state. These confined spaces are shown to behave as host units in the solid state for guests including solvent molecules and charge balancing counter anions. Significant enhancements in enantioselectivities and reaction efficiencies in asymmetric copper-catalysed C-H insertion and aromatic addition reactions of α-diazocarbonyl compounds in the presence of various group I salts are reported. Forman's discrete Morse theory is studied from an algebraic viewpoint, and we show how this theory can be extended to chain complexes of modules over arbitrary rings. As applications we compute the homologies of a certain family of nilpotent Lie algebras, and show how the algebraic Morse theory can be used to derive the classical Anick resolution as well as a new two-sided Anick resolution.

Abstract:
Superoxide (O-2(-): IUPAC name dioxide (center dot 1-)) is an important transient reactive oxygen species (ROS) in the ocean formed as an intermediate in the redox transformation of oxygen (O-2) into hydrogen peroxide (H2O2) and vice versa. This highly reactive and very short-lived radical anion can be produced both via photochemical and biological processes in the ocean. In this paper we examine the decomposition rate of O-2(-) throughout the water column, using new data collected in the Eastern Tropical North Atlantic (ETNA) Ocean. For this approach we applied a semi factorial experimental design to identify and quantify the pathways of the major identified sinks in the ocean. Abstract:
While protein-protein interactions have been studied largely as a network graph without physicality, here we analyze two protein complex data sets of Saccharomyces cerevisiae to relate physical and functional modularity to the network topology. We study for the first time the number of different protein complexes as a function of the protein complex size and find that it follows an exponential decay with a characteristic number of about 7. This reflects the dynamics of complex formation and dissociation in the cell. The analysis of the protein usage by complexes shows an extensive sharing of subunits that is due to the particular organization of the proteome into physical complexes and functional modules. This promiscuity accounts for the high clustering in the protein network graph. Our results underscore the need to include the information contained in observed protein complexes into protein network analyses.

Abstract:
The synthesis, structures and magnetic characterisation of a family of discrete planar pentanuclear Cu(II) 12-MC-4 metallacrowns of formulae [Cu-5(L-1)(4)(MeOH)(4)](ClO4)(2) (1), [Cu-5(L-1)(4)(py)(2)](ClO4)(2)center dot py (2), [Cu-5(L-1)(4)(py)(6)](ClO4)(2) (3) and [Cu-5(L-2)(4)(MeOH)(4)](ClO4)(2)center dot H2O (7) (where L1H2 = 2-(dimethylamino)phenylhydroxamic acid and L2H2 = 2-(amino)phenylhydroxamic acid) are reported. UV-vis and Electrospray MS studies indicate solution stability with respect to their {Cu-5(L)(4)}(2+) cores. Magnetic susceptibility measurements confirm strong antiferromagnetic exchange between the Cu(II) ions resulting in isolated S = 1/2 ground spin states. Abstract:
Objective. Tumor necrosis factor receptor (TNFR) -associated periodic syndrome (TRAPS) is an autosomal-dominant autoinflammatory condition caused by mutations in the TNFRSFIA gene. Unlike other autoinflammatory diseases in which anti-TNF therapy is largely a successful treatment option, therapy with the anti-TNF drug infliximab is often ineffective in patients with TRAPS. Moreover, in certain cases, infliximab actually triggers severe episodes of inflammation. The aim of this study was to elucidate the mechanisms underlying such a reaction. Methods. Peripheral blood mononuclear cells (PBMCs) were obtained from patients with TRAPS. Both caspase 3 activity and NF-kappa B subunit activity were determined by enzyme-linked immunosorbent assay. Cytokine secretion was assessed using a specific customized human multiplex bead immunoassay kit. Results.
