Abstract:
Chronic allograft nephropathy (CAN) remains the leading cause of renal graft loss after the first year following renal transplantation. This study aimed to identify novel urinary proteomic profiles, which could distinguish and predict CAN in susceptible individuals. Experimental Design: The study included 34 renal transplant patients with histologically proven CAN and 36 patients with normal renal transplant function. High-throughput proteomic profiles were generated from urine samples with three different ProteinChip arrays by surface-enhanced laser-desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS). Following SELDI a biomarker pattern software analysis was performed which led to the identification of a novel biomarker pattern that could distinguish patients with CAN from those with normal renal function. Results: An 11.7 kDa protein identified as β2 microglobulin was the primary protein of this biomarker pattern, distinguishing CAN from control patients (ROC =...

Abstract:
We report on the use of Zn as an n-type dopant in CuCl thin films for optoelectronic applications, wherein maximum n-type doping of the order of 1018 cm -3 has been achieved. Zn doped nanocrystalline CuCl thin films are successfully deposited on glass and Si substrates by pulsed dc magnetron sputtering. Structural and morphological properties are investigated using X-ray diffraction (XRD) studies and Scanning Electron Microscopy (SEM), respectively. The conductivity of the CuCl:Zn films is examined using the four point probe technique. An order of magnitude increase in the conductivity of CuCl, by the doping with Zn is reported herein. The doped CuCl films display strong room temperature cathodoluminescence (CL) at ~ 385nm, which is similar to that of the undoped films. Hall Effect measurements show an n-type conductivity of the doped films.

Abstract:
The genesis of hydrothermal ore deposits is of crucial economic importance. This study investigates the extent, causes and consequences of zinc and iron isotope fractionation in a large hydrothermal system at the world-class Navan Zn–Pb orebody, Ireland. Large variations in Zn, Fe and S isotope compositions have been measured in microdrilled sphalerite (ZnS) at the millimetre scale. d66Zn and d56Fe display a well-defined positive correlation and both also correlate with d34S. These relationships represent the combined effects of kinetic Zn and Fe isotope fractionation during sphalerite precipitation, and S isotope variation through mixing of hot, metal-rich hydrothermal fluids and cool, bacteriogenic sulfide-bearing brines. Combined with S isotope data, d56Fe and d66Zn data on mine concentrates confirm that hydrothermal sulfide is a minor component of the overall deposit signature. Our data suggest that incoming pulses of metal-rich hydrothermal fluid triggered sulfide mineralisatio...

Abstract:
ZNF804A rs1344706 is the first genetic risk variant to achieve genome wide significance for psychosis. Following earlier evidence that patients carrying the ZNF804A risk allele had relatively spared memory function compared to patient non-carriers, we investigated whether ZNF804A was also associated with variation in brain volume. In a sample of 70 patients and 38 healthy participants we used voxel based morphometry to compare homozygous (AA) carriers of the ZNF804A risk allele to heterozygous and homozygous (AC/CC) non-carriers for both whole brain volume and specific regions implicated in earlier ZNF804A studies ?the dorsolateral pre-frontal cortex, the hippocampus, and the amygdala. For patients, but not for controls, we found that homozygous `AA? risk carriers had relatively larger gray matter volumes than heterozygous/homozygous non-carriers (AC/CC), particularly for hippocampal volumes. These data are consistent with our earlier behavioral data and suggest that ZNF804A is deli...

Abstract:
ZnO nanorods grown by both high temperature vapour phase transport and low temperature chemical bath deposition are very promising sources for UV third harmonic generation. Material grown by both methods show comparable efficiencies, in both cases an order of magnitude higher than surface third harmonic generation at the quartz-air interface of a bare quartz substrate. This result is in stark contrast to the linear optical properties of ZnO nanorods grown by these two methods, which show vastly different PL efficiencies. The third harmonic generated signal is analysed using intensity dependent measurements and interferometric frequency resolved optical gating, allowing extraction of the laser pulse parameters. The comparable levels of efficiency of ZnO grown by these very different methods as sources for third harmonic UV generation provides a broad suite of possible growth methods to suit various substrates, coverage and scalability requirements. Potential application areas range f...

Abstract:
We correct Eq. (1) of our previous paper showing the pulse duration (τ) dependence on group delay dispersion (β2) and initial duration of the unchirped pulse coming from the laser (τ0). The correct form of the equation was used in all the calculations in the previous paper however, so in all other respects our previous results and conclusions are unchanged.

Zoochorous dispersal of freshwater bivalves: an overlooked vector in biological invasions?
(2017)

Abstract:
Vectors that underpin the natural dispersal of invasive alien species are frequently unknown. In particular, the passive dispersal (zoochory) of one organism (or propagule) by another, usually more mobile animal, remains poorly understood. Field observations of the adherence of invasive freshwater bivalves to other organisms have prompted us to assess the importance of zoochory in the spread of three prolific invaders: zebra mussel Dreissena polymorpha; quagga mussel Dreissena bugensis; and Asian clam Corbicula fluminea. An extensive, systematic search of the literature was conducted across multiple on-line scientific databases using various search terms and associated synonyms. In total, only five publications fully satisfied the search criteria. It appears that some fish species can internally transport viable adult D. polymorpha and C. fluminea specimens. Additionally, literature indicates that veligers and juvenile D. polymorpha can adhere to the external surfaces of waterbirds....

Zooming Algorithms for Digital Holography
(2010)
Hennelly, Bryan M.; Kelly, Damien; Pandey, Nitesh; Monaghan, David
Abstract:
Digital Holography is an imaging modality made up of two parts: (i) Recording an interference pattern on a CCD, where an object wave field and a known reference wave are coincident and extracting the complex object wave field from this interference pattern and (ii) Replaying or reconstructing the hologram on a computer by simulating the propagation of the object wave field back to the object plane. Thus an image is obtained. We show how to adapt the reconstruction algorithm in a simple way to allow it to generate any output range and in any location making it far more versatile for zooming in on specific regions of our reconstructed image.

ZPP is Hard Unless RP is Small

Abstract:
We use Lutz's resource bounded measure theory to prove that, either RP is small, or ZPP is hard. More precisely, we prove that if RP has not p-measure zero, then EXP equals ZPP on infinitely many input lengths, i.e. there are infinitely many input lengths on which ZPP is hard. Second we prove that if NP has not p-measure zero, then derandomization of AM is possible on infinitely many input length, i.e. there are infinitely many input lengths such that NP = AM. Finally we prove easiness versus randomness tradeoffs for classes in the polynomial time hierarchy. We show that it appears to every strong adversary that either, every Ʃᴾᵢ algorithm can be simulated infinitely often by a subexponential co-nondeterministic time algorithm, having oracle access to Ʃᴾᵢ -2 , or BP Ʃᴾᵢ = Ʃᴾᵢ .

Abstract:

Abstract:
Hereditary deficiency of the protein α-1 antitrypsin (AAT) causes a chronic lung disease in humans that is characterized by excessive mobilization of neutrophils into the lung. However, the reason for the increased neutrophil burden has not been fully elucidated. In this study we have demonstrated using human neutrophils that serum AAT coordinates both CXCR1- and soluble immune complex (sIC) receptor-mediated chemotaxis by divergent pathways. We demonstrated that glycosylated AAT can bind to IL-8 (a ligand for CXCR1) and that AAT-IL-8 complex formation prevented IL-8 interaction with CXCR1. Second, AAT modulated neutrophil chemotaxis in response to sIC by controlling membrane expression of the glycosylphosphatidylinositol-anchored (GPI-anchored) Fc receptor FcγRIIIb. This process was mediated through inhibition of ADAM-17 enzymatic activity. Neutrophils isolated from clinically stable AAT-deficient patients were characterized by low membrane expression of FcγRIIIb and increased chem...

α-Keto amides as precursors to heterocycles—generation and cycloaddition reactions of piperazin-5-one nitrones

Abstract:
α-Keto amides 10a,b, formed from reaction of pyruvic or benzoylformic acid with allyl amine are found to present as single rotameric forms whilst their tertiary amido analogues 10c,d present as two rotamers in solution at rt. The hydroxyimino derivatives 8 share the conformational characteristics of their parents. The geometrical make-up of the new α-amidooximes is seen to depend on the structure of the starting acid and on the degree of substitution of the amido group. The oxime 8a derived from pyruvic acid and allyl amine is formed solely as the (E)-isomer whilst its tertiary amido analogue 8c is formed as both (E)- and (Z)-isomers. Oximes derived from benzoylformic acid have the opposite selectivity with both geometrical isomers forming from the secondary amide 8b and only the (Z)-isomer from the tertiary amide 8d. With the exception of 8b all oximes were configurationally stable with (Z)-isomers reacting to form isoxazolopyrrolidinones 11—compounds with a relatively rare bicycli...

Abstract:
Preparation of a series of terminally and internally substituted δ-alkenyl and δ-alkynyl esters 6,7 and 9, potential precursors to oxazin-2-one nitrones, has been attempted. Condensation between pyruvic or benzoylformic acid and the appropriate alcohol proceeded smoothly in some cases whilst allylic transposition was a major feature in other cases – most especially during reactions with α-vinylbenzyl alcohol. Oximation of pyruvic acid derivatives furnished E-oxime isomers whilst benzoylformic acid derivatives afforded mixed geometrical isomers. The E-oxime of 4a1 carrying an internal Me group undergoes facile thermal cyclisation affording nitrones 1c and 1d in good yield. Oximes E-5a,b with a terminal methyl substituent on the alkene moiety furnish nitrone only under the influence of an external electrophile [PhSeBr/AgBF4]. A terminal Ph substituent on 5c,d prohibits formation of the cyclic dipole irrespective of reaction conditions, and whilst 5d reacts to afford a bicyclic isoxazo...

Abstract:
<p>Twelve novel β-lactams were synthesised and their antiproliferative effects and binding affinity for the predominant isoforms of the estrogen receptor (ER), ERα and ERβ, were determined. β-Lactams <strong>23 </strong>and <strong>26</strong> had the strongest binding affinities for ERα (IC50 values: 40 and 8 nM respectively) and ERβ (IC50 values: 19 and 15 nM). β-Lactam <strong>26 </strong>was the most potent in antiproliferative assays using MCF-7 breast cancer cells, and further biochemical analysis showed that it caused accumulation of cells in G2/M phase (mitotic blockade) and depolymerisation of tubulin in MCF-7 cells. Compound <strong>26</strong> also induced apoptosis and downregulation of the pro-survival proteins Bcl-2 and Mcl-1. Computational modeling predicted binding preferences for the dual ER/tubulin ligand <strong>26</strong>. This s...

Abstract:
<p>The original article is available at http://pubs.rsc.org/en/content/articlelanding/2012/ra/c2ra01351g#!divAbstract</p>
<p>The first hybrid molecule of a <em>β</em>-lactam antibiotic and a host defence peptide and a method for the preparation of this type of molecule are reported. Conjugation of an antimicrobial peptide to a cephalosporin, through a cleavable linker, reversibly masks one of the activity determinants of the peptide. Its release from the <em>β</em>-lactam core can be selectively triggered by bacterial enzymes (<em>β</em>-lactamases) which mediate resistance to <em>β</em>-lactam agents. A prototypical cephalothin-bactenecin candidate was synthesised, using a copper(I)-catalysed azide–alkyne cyclo-addition reaction for the conjugationstep. Enzymatic hydrolysis assays of this candidate were initially performed with a purified <em>β</em>-lactamase to confirm that the peptide can be release...

The dairy protein β-lactoglobulin (βlg) is known to bind hydrophobic ligands such as fatty acids. In the present work, we investigated the biological activity in vitro of linoleate once complexed to bovine βlg. Binding of linoleate (C18:2) to bovine βlg was achieved by heating at 60 °C for 30 min at pH 7.4, resulting in a linoleate/βlg molar binding stoichiometry of 1.1, 2.1, and 3.4. Two types of binding sites were determined by ITC titrations. Binding of linoleate induced the formation of covalent dimers and trimers of βlg. The LD50 on Caco-2 cells after 24 h was 58 μM linoleate. However, cell viability was unaffected when 200 μM linoleate was presented to the Caco-2 cells as part of the βlg complex. The Caco-2 cells did not increase mRNA transcript levels of long chai...

The dairy protein β-lactoglobulin (BLG) is known to bind fatty acids such as the salt of the essential longchain fatty acid linoleic acid (cis,cis-9,12-octadecadienoic acid, n-6, 18:2). The aim of the current study was to investigate how bovine BLG-linoleate complexes, of various stoichiometry, affect the enzymatic digestion of BLG and the intracellular transport of linoleate into enterocyte-like monolayers. Duodenal and gastric digestions of the complexes indicated that BLG was hydrolyzed more rapidly when complexed with linoleate. Digested as well as undigested BLG-linoleate complexes reduced intracellular linoleate transport as compared with free linoleate. To investigate whether enteroendocrine cells perceive linoleate differently when part of a complex, the ability ...
